Most vacancies don't specify maybe the cover leter should be in msword format as attachment, hand written and attached or just typed as the body of the application.

If it online submission it should be in MS Word or pdf if specified and u don't need to sign (your name will take care of the signature)

I don't think you really need to sign a cover letter. Your name will just do.

However, it is always advisable to have your cover letter and cv/resume on pages simultaneously. That is to say if your cv/resume is a one or two paged document as the case maybe, then your cover letter should be page1 while your cv/resume becomes page2... 3... 4...

Most Job applications this days are sent through email, in which case the cover letter forms the body of the email, (no one would want to download your application letter before reading it), professional ATS site do specifically ask for a cover letter, in this case you upload it as a document just like your resume.
