I'm very interested in the development of Vaccine and treatment of Ebola.


Garcinia kola also known as bitter Kola has shown some promise. Experts in pharmacognosy can shed more light on this.

Survivors of Ebola are confered permanent immunity. Scientists, Serologists and Immunologists can shed more light on how this happens.

Can an assymptomatic patient transmit
the virus during incubation period?...Epidemiologists should tell us what they think.

Can the Serum of a survivor really help in the treatment of infected patient?...if true..how does this happens?

What other Strains of Ebola do we have and which is more Virulent?

The general principle of most vaccination is the administration of attenuated doses of the DISEASE-CAUSING organism (e.g Ebola) to elicit antibody-antigen reaction and produce Memory T-cells which will help the body 'remember' not to get infected next time the organism tries to invade the body..

How does this help in the Ebola issue?. How do we attenuate the Ebola virus?
Can we identify the part of the virus that causes virulence and mortality in humans and then 'delete' before introduction as vaccines.

As the following article says...

Antibody titers against Ebola virus GPs are readily detectable in patients who recover from Ebola virus infection; however, anecdotal reports have indicated that serum from recovered patients did not consistently protect against infection or exhibit neutralization of virus replication in cell culture. Furthermore, passive transfer of antibodies in animal models only delays the onset of symptoms and does not alter overall survival . More recently, the neutralization of virus replication by selected monoclonal antibodies isolated from the bone marrow of recovered patients was demonstrated in vitro and monoclonal antibodies that recognize specific epitopes of Ebola virus GP have been shown to confer immune protection in a murine model of Ebola virus infection (15, 45) and in guinea pigs (25). However, it is relatively easy to protect against infection in the mouse model, and protection of guinea pigs required a high dose of antibody administered very close to the time of virus challenge. Taken together, these results suggest that antibodies alone do not provide protective immunity in a natural context and that cellular immunity is likely to play a significant role in virus clearance. Whether hyperimmune serum from surviving vaccinated animals or certain infrequently occurring antibodies are capable of attenuating infection remains unknown, but such antibodies could potentially contribute to therapy if they can be identified and optimized.

In other words...the serum method might work....but it remains to be seen.

The article quoted also appears to suggest(based on a study done in Gabon) that a initial immune response that is T-cell based is crucial to surviving Ebola.

Survivors exhibited more significant IgM responses, clearance of viral antigen, and sustained T-cell cytokine responses, as indicated by high levels of T-cell-related mRNA in the peripheral blood. In contrast, antibodies specific for the virus were nearly undetectable in fatal cases, and while gamma interferon (IFN-γ) was detected early after infection, T-cell cytokine RNA levels were more indicative of a failure to develop adaptive immunity in the days preceding death.

Thank you very much for submission i followed your links they were intellectually juicy. Something caught my interest on the Gabon study link -the immunologic responses of survivors in contrast to ebola fatality cases.

In survivors, early and increasing levels of IgG, directed mainly against the nucleoprotein and the 40-kDa viral protein, were followed by clearance of circulating viral antigen and activation of cytotoxic T cells, which was indicated by the upregulation of FasL, perforin, CD28 and gamma interferon mRNA in peripheral blood mononuclear cells. In contrast, fatal infection was characterized by impaired humoral responses, with absent specific IgG and barely detectable IgM.

From above increasing levels of IgG seems to be key for survival, we also know about Intravenous Immunoglobulin.

This question begs-Can IgG be transfused in this case as the patient's immune system battles to mop up the invasion of the virus?

We may just try it out on a patient in controlled high-level biosafety milieus. But timing will be key as "events very early in Ebola virus infection determine the control of viral replication and recovery or catastrophic illness and death", if i may quote the same source.

Back to topic...

I read on Yahoo news that one of the American medics Dr. Kent, asked for transfusion of blood from the a little boy he treated. That seem to substantiate the claim that IgG antibodies specifically raised from antibody-antigen reaction during ebola invasion could be helpful in the treated of new Ebola cases as i previously opined.

Prof Iwu a Virologist also talked about the invitro anti-ebola actions of Garcinia Kola, but admitted that the chemical structure maybe altered to give sustainable results invivo. Structural and pharmaceutical Chemists should shed more light on this...
