Mpg isn't calculated only by engine cylinder other variables are involved

It depends on the car model in question ,kindly note that some 4 plugs consume more than some 6 plugs .

Below are the fuel consumption figures for an RX 300 according to Lexus.

City Driving: 16mpg
Highway Driving : 21mpg

A 2003 model year 4-Cylinder RAV4 will do the following according to Toyota

City Driving: 21mpg
Highway Driving : 27mpg

A 2005 model year 4-Cylinder Honda EOD will do the following according to Honda

City Driving: 21mpg
Highway Driving : 31mpg

So your decision will really be influenced by the car you are comparing the RX300 to. Please note that these are the figures touted by the manufacturers when these cars are brand new.

Real life figures will be different especially as we incorporate all the other factors - age, state of engine, etc. And also that Lagos state has a different category not captured by any of the manufacturers which is traffic / go-slow driving.

OP all other things being equal a V6 will consume more fuel than an i4 of same model, same manufacturer, same technology. But d V6 variant will have more power, more torque and more agility when compared with its i4 sibling.
Case in point a 2007 E350 (V6) vs 2007 E200 (i4).

For your Lexus though RX300 doesn't come in other variants than the 3.0L V6 unless u wanna get Harrier which doesn't carry the Lexus badge.
Fuel consumption is ok for that particular Lexus and it's a soft ride to drive, just don't allow one Kazeem remove ur thermostat, cos that will go a long way in tampering with fuel economy.

depends mostly on the cylinder size/bore( ratio of top dead centre to bottom dead centre).A four piston engine having a large cylinder would consume more than the standard small bore v6 engine.

My Car is 12plugs.
The model of my car also has a 4plugs version, ironically my friend who has d 4plugs version complains of High fuel consumption.

There are so many factors dat determines fuel consumption.
Pattern of driving.
age of engine(not age of car)
Maintenance
Type of fuel used.
Tyre pressure n type of tyres used
E. T. C

Other factors are also involved.
A 6 cylinder engine, may still save more gas than an I4 vehicle, when there's air leakage from the exhaust, bad injector and bad O2 sensor

my dear, am using a toyota Avalon 2004 its a V6 Engine. its relatively ok. lexus is an extension of toyota so it should also be economical. just google it.

Do you know the Aerodynamics effect on a vehicle, whose windows are wound down and it is at a top speed let's say 130km/h? It is more like racing on a wide wind with a deployed parachute. The OP is asking how much will it consume on a journey; underline that. If you guys have settle down to read your vehicle utility manual, you would understand that the manufacturer gave reasons why you must wind up your side windows when traveling on highway with your HVAC system to save fuel.
If I can use 37ltrs of fuel from Lagos to Benin on a 3.5L V6 2002 Infiniti QX4 heavy, with AC ON I do not see the argument.

Forget fuel consumption and get your car.
I owe a lexus ls460 it's a v8 engine that's how people were using panadol for my headache, shouting it consumes fuel maintenance and all. I think decide what you want and for what purpose and go for it.

My take you will enjoy a v8/v6 more while travelling compare to a 4 plug engine, and you will thank your stars for having a 4 plug driving in lagos holdups and during fuel scarcity

Lexus 300 is a suv with comfortable/roomy interior & high ground clearance for nigerian roads. I have a highlander. A V6 will only consume more fuel if you decide to drive at high speeds always.
