Currently there's no effective defense against such missile. Mind you a missile defense is divided into two :
Soft kill and hard kill/kinetic defense.

Soft kill requires the use of decoys, radar jamming or electronic spoofing. Its used to make sure the missile missed its intended target by interfering with missile targeting systems and flight paths.

Hard kill requires the use of kinetic force. Through an explosive warhead or sheer kinetic force. It intercept the missile at one of its different flight phase and causes total or partial destruction of missile system.

Currently the only defence US have against hypersonic missile is the soft kill system which itself has limited effectiveness. There's no hard kill system currently or in development that can defeat this Russia weapon. Mind you even against mere supersonic missile the most advanced air defense system can not guarantee a 100% interception rate even 80% is term a success. So missile defense isn't as easy as you think
