Health › AZD1222 vaccine met primary efficacy endpoint in preventing COVID-19 by References(op): 8:34am On Nov 23, 2020
Two different dosing regimens demonstrated efficacy with one showing a better profile

No hospitalisations or severe cases of COVID-19 in participants treated with AZD1222

Positive high-level results from an interim analysis of clinical trials of AZD1222 in the UK and Brazil showed the vaccine was highly effective in preventing COVID-19, the primary endpoint, and no hospitalisations or severe cases of the disease were reported in participants receiving the vaccine. There were a total of 131 COVID-19 cases in the interim analysis.

One dosing regimen (n=2,741) showed vaccine efficacy of 90% when AZD1222 was given as a half dose, followed by a full dose at least one month apart, and another dosing regimen (n=8,895) showed 62% efficacy when given as two full doses at least one month apart. The combined analysis from both dosing regimens (n=11,636) resulted in an average efficacy of 70%. All results were statistically significant (p<=0.0001). More data will continue to accumulate and additional analysis will be conducted, refining the efficacy reading and establishing the duration of protection.

An independent Data Safety Monitoring Board determined that the analysis met its primary endpoint showing protection from COVID-19 occurring 14 days or more after receiving two doses of the vaccine. No serious safety events related to the vaccine have been confirmed. AZD1222 was well tolerated across both dosing regimens.

AstraZeneca will now immediately prepare regulatory submission of the data to authorities around the world that have a framework in place for conditional or early approval. The Company will seek an Emergency Use Listing from the World Health Organization for an accelerated pathway to vaccine availability in low-income countries. In parallel, the full analysis of the interim results is being submitted for publication in a peer-reviewed journal.

Professor Andrew Pollard, Chief Investigator of the Oxford Vaccine Trial at Oxford, said: “These findings show that we have an effective vaccine that will save many lives. Excitingly, we’ve found that one of our dosing regimens may be around 90% effective and if this dosing regime is used, more people could be vaccinated with planned vaccine supply. Today’s announcement is only possible thanks to the many volunteers in our trial, and the hard working and talented team of researchers based around the world.”

Pascal Soriot, Chief Executive Officer, said: “Today marks an important milestone in our fight against the pandemic. This vaccine’s efficacy and safety confirm that it will be highly effective against COVID-19 and will have an immediate impact on this public health emergency. Furthermore, the vaccine’s simple supply chain and our no-profit pledge and commitment to broad, equitable and timely access means it will be affordable and globally available, supplying hundreds of millions of doses on approval.”

The pooled analysis included data from the COV002 Phase II/III trial in the UK and COV003 Phase III trial in Brazil. Over 23,000 participants are being assessed following two doses of either a half-dose/full-dose regimen or a regimen of two full doses of AZD1222 or a comparator, meningococcal conjugate vaccine called MenACWY or saline. The global trials are evaluating participants aged 18 years or over from diverse racial and geographic groups who are healthy or have stable underlying medical conditions.

Clinical trials are also being conducted in the US, Japan, Russia, South Africa, Kenya and Latin America with planned trials in other European and Asian countries. In total, the Company expects to enrol up to 60,000 participants globally.

The Company is making rapid progress in manufacturing with a capacity of up to 3 billion doses of the vaccine in 2021 on a rolling basis, pending regulatory approval. The vaccine can be stored, transported and handled at normal refrigerated conditions (2-8 degrees Celsius/ 36-46 degrees Fahrenheit) for at least six months and administered within existing healthcare settings.

AstraZeneca continues to engage with governments, multilateral organisations and collaborators around the world to ensure broad and equitable access to the vaccine at no profit for the duration of the pandemic.

COV002
COV002 is a single-blinded, multi-centre, randomised, controlled Phase II/III trial assessing the safety, efficacy and immunogenicity of AZD1222 in 12,390 participants in the UK. Trial participants to date are aged 18 years or over, who are healthy or have medically stable chronic diseases and are at increased risk for being exposed to the SARS-CoV-2 virus. Participants receive one or two intramuscular doses of a half dose (~2.5 x1010 viral particles) or full dose (~5x1010 viral particles) of AZD1222 or comparator, meningococcal vaccine MenACWY. Participants have blood samples drawn and clinical assessments for safety as well as immunogenicity at multiple timepoints up to one year post-vaccination. Suspected cases presenting with compatible symptoms were tested for virological confirmation by COVID-19 PCR. In addition, weekly swabbing are done for detection of infection and assessment of vaccine efficacy against infection.

COV003
COV003 is a single-blinded, multi-centre, randomised, controlled Phase III trial assessing the safety, efficacy, and immunogenicity of AZD1222 in 10,300 participants in Brazil. Trial participants to date are aged 18 years or over, who are healthy or have medically stable chronic diseases and are at increased risk for being exposed to the SARS-CoV-2 virus. Participants are randomised to receive two intramuscular doses of a full dose (~5x1010 viral particles) of AZD1222 or comparator, meningococcal vaccine MenACWY as first dose and a saline placebo as second dose. Participants have blood samples drawn and clinical assessments for safety as well as immunogenicity at multiple timepoints up to one year post-vaccination. Suspected cases presenting with compatible symptoms were tested for virological confirmation by COVID-19 PCR.

AZD1222
AZD1222 was co-invented by the University of Oxford and its spin-out company, Vaccitech. It uses a replication-deficient chimpanzee viral vector based on a weakened version of a common cold virus (adenovirus) that causes infections in chimpanzees and contains the genetic material of the SARS-CoV-2 virus spike protein. After vaccination, the surface spike protein is produced, priming the immune system to attack the SARS-CoV-2 virus if it later infects the body.

Politics › Sylva: COVID-19 Vaccine Discovery, Remote Cause Of Petrol Price Increase by References(op): 6:41pm On Nov 16, 2020
The recent upwards increase in petrol pump price, from N158 to N168, has been attributed to the announcement of a possibility of having a vaccine for the Novel Coronavirus (COVID-19) Pandemic soon.

Minister of State for Petroleum Resources, Timipre Sylva, disclosed this while addressing State House Correspondents on Monday, after briefing President Muhammadu Buhari in his office at the Aso Rock Presidential Villa in Abuja.

The Minister also assured that Nigerians would get used to the deregulation of the pump price petrol just as they have become the case of kerosene and diesel.

According to him, diesel and kerosene are even more important to the ordinary citizens than petrol, which he said is mostly used by the elite.

He explained that trucks that move food produce from one part of the country to another use diesel while kerosene also used by most of the Nigerian masses has since been deregulated.

He said: “Look at it, a situation where diesel has been deregulated long ago, a situation where kerosene has been deregulated long ago, and these are what the poorest people in Nigeria interact with more. Why do I say that?

“If you want to transport food from the north to the south, it will be by trucks that are run by diesel, not with petrol. Those trucks that transport food from the north to the south are usually run by diesel.

“Kerosene is the preferred fuel at the lowest level of our society. These have been deregulated long ago. So, what is the problem with deregulating petrol, which is mostly used by the elite?

“Let us be fair to this country, let us be fair to the poor people in this country.

“If we have deregulated what they were using, then there is actually no reason why we should continue to subsidize petrol. I feel so. That’s my personal feeling.”

NYSC › Two Corpers Test Positive For COVID-19 In Kano by References(op): 3:35pm On Nov 16, 2020
The NYSC Coordinator in Kano State, Hajiya Aisha Tata, says two new corps members out of the 826 posted to the state have tested positive for COVID-19.

The coordinator disclosed this on Monday while briefing newsmen after the swearing-in of the 2020 Batch ‘B’ Stream A corps members.

She said the corps members were tested immediately upon arrival at the orientation camp and that they were taken to relevant facilities for further medical investigation.

She said that 826 corps members posted to the state have undergone screening on COVID-19 before they were allowed access to the camp promises.

“As you have seen, we have provided all the necessary prevention facilities and ensured that all the corps members used them.

“We have also ensured comprehensive fumigation to ensure that the camp remains a clean environment for the corps members,’’ she said.

In his message to the ceremony, Governor Abdullahi Ganduje called on the corps members to remain calm, and participate in all activities with open minds.

“My fellow compatriots, the orientation course was purposely designed to provide you with the requisite training for a better understanding and knowledge about the scheme, about your new environment and about the people you are going to be staying with after the orientation course.

“I implore you to be guided by the sense of duty and discipline that saw you through your various academic programmes.

“You are enjoined to shun all acts that may jeopardise harmonious relationships existing between the corps members and their host communities,” he said.

He reiterated his administration’s commitment to continue to identify with NYSC programmes and support it to realise the purpose for which it was set up.

Ganduje assured the corps members of their security, safety and welfare throughout their one- year stay in the state.

The governor was represented by the Permanent Secretary, Kano State Ministry of Youths and Sports Development, Alhaji Ibrahim Ahmed.

News Agency of Nigeria reports that the oath of allegiance was administered on the corps members by Chief Magistrate Jibril Muhammad.

Of the 826 corps members deployed in Kano State, 387 are females, while 439 are males.

Something extremely bogus is going on. Was tested for covid four times today. Two tests came back negative, two came back positive. Same machine, same test, same nurse. Rapid antigen test from BD.

Foreign Affairs › Ukrainian President, Chief of Staff, Finance Sec test Positive For COVID-19 by References(op):
Ukrainian President Volodymyr Zelenskiy and two top presidential and government officials have tested positive for the coronavirus, presidential and government offices said on Monday.

"The head of state is feeling well and will continue to perform his duties remotely in self-isolation," Zelenskiy's office said in a statement.

"Despite all the quarantine measures, I also received a + (positive) result. I have 37.5, and I wish everyone 36.6!" Zelenskiy said in a Telegram message, referring to his body temperature.

Zelenskiy's wife, Olena, contracted COVID-19 in June and spent several weeks in a hospital.

Minutes after Zelenskiy's announcement, Andriy Yermak, head of the presidential office, said on Facebook that he had also tested positive for the virus.

Finance Minister Serhiy Marchenko has been diagnosed with COVID-19 as well and will work remotely, his ministry said.

On Thursday, Marchenko, Prime Minister Denys Shmygal and other ministers took part in a parliamentary session. Shmygal's office said he had no signs of the infection but would undergo a test on Tuesday.

The daily tally of coronavirus infections in Ukraine spiked in late September and remained consistently high throughout October and early November, prompting the government to extend lockdown measures until the end of this year.

Shmygal said last week the number of new cases could jump to 15,000 a day by the end of November and to 20,000 daily by the end of the year.

Ukraine's health minister said last week the coronavirus situation in Ukraine was close to catastrophic and that the nation must prepare for the worst.

Zelenskiy said earlier on Monday Ukraine may introduce a lockdown at weekends in an effort to curb the pandemic and such a move would not have a serious negative impact on the economy.

There are no lucky people for whom #COVID19 does not pose a threat. Despite all the quarantine measures, I received a positive test. I feel good & take a lot of vitamins. Promise to isolate myself, but keep working. I will overcome COVID19 as most people do. It's gonna be fine!
