Health / New Software Can Verify Someone's Identity By Their DNA In Minutes by gabonsky: 6:49pm On Dec 01, 2017
Researcher Sophie Zaaijer uses the MinION, a portable DNA sequencer, to get a quick genetic readout of a sample of cells.


In the science-fiction movie Gattaca, visitors only clear security if a blood test and readout of their genetic profile matches the sample on file. Now, cheap DNA sequencers and custom software could make real-time DNA-authentication a reality.


Researchers at Columbia University and the New York Genome Center have developed a method to quickly and accurately identify people and cell lines from their DNA. The technology could have multiple applications, from identifying victims in a mass disaster to analyzing crime scenes. But its most immediate use could be to flag mislabeled or contaminated cell lines in cancer experiments, a major reason that studies are later invalidated. The discovery is described in the latest issue of eLife.
"Our method opens up new ways to use off-the-shelf technology to benefit society," said the study's senior author Yaniv Erlich, a computer science professor at Columbia Engineering, an adjunct core member at NYGC, and a member of Columbia's Data Science Institute. "We're especially excited about the potential to improve cell-authentication in cancer research and potentially speed up the discovery of new treatments."


The software is designed to run on the MinION, an instrument the size of a credit card that pulls in strands of DNA through its microscopic pores and reads out sequences of nucleotides, or the DNA letters A, T, C, G. The device has made it possible for researchers to study bacteria and viruses in the field, but its high error-rate and large sequencing gaps have, until now, limited its use on human cells with their billions of nucleotides.


In an innovative two-step process, the researchers outline a new way to use the $1,000 MinION and the abundance of human genetic data now online to validate the identity of people and cells by their DNA with near-perfect accuracy. First, they use the MinION to sequence random strings of DNA, from which they select individual variants, which are nucleotides that vary from person to person and make them unique. Then, they use a Bayesian algorithm to randomly compare this mix of variants with corresponding variants in other genetic profiles on file. With each cross-check, the algorithm updates the likelihood of finding a match, rapidly narrowing the search.


Tests show the method can validate an individual's identity after cross-checking between 60 and 300 variants, the researchers report. Within minutes, it verified the identity of the study's lead author, Sophie Zaaijer, a former member of NYGC and now a postdoctoral researcher at Cornell Tech.
To do this, the MinION matched the readout of Zaaijer's genome, gleaned from a sample of cheek cells, with a reference profile stored among 31,000 other genomes on the public database, DNA.land. Erlich's identity was verified the same way, with initial sequencing done by Columbia students in the Ubiquitous Genomics class he and Zaaijer taught in 2015.


They call their re-identification technique 'MinION sketching' which Zaaijer compares to the brain's ability to make out a bird from a few telling features in an abstract Picasso line-drawing. The MinION's genetic 'sketch' of a cell-sample is compared to a growing database of sketches -- similarly incomplete genetic profiles produced by at-home DNA-test kits like 23andMe and donated to science by consumers.


"Using our method, one needs only a few DNA reads to infer a match to an individual in the database," says Zaaijer.


The most promising use for 'MinION sketching' may be as a cheap cell-authentication tool in experimental research, say scientists familiar with its capabilities. In the study, researchers quickly matched a strain of leukemic cells sequenced by the MinION against a reference file in the Cancer Cell Line Encyclopedia database, they report. When they tried contaminating the cells with other cultures, it correctly rejected a match if contamination levels climbed above 25 percent.


The use of misidentified or contaminated cell lines in medical research is blamed for as much as a third of the estimated $28 billion spent each year on studies that can't be replicated, according to one recent study. In a 2014 essay in Science, the director of the National Institute of General Medical Sciences, Jon Lorsch, called for new policies and technologies to address the problem.


Lacking the expensive machinery needed to validate cell lines on their own, most researchers either skip validation or ship their cultures to specialized labs which can delay important findings and treatments. If an easier alternative were available, most researchers would use it, says Neville Sanjana, a core faculty member at NYGC and assistant professor at NYU's Department of Biology who works on skin and lung cancer cell lines and was not involved in the study.


"No one wants to waste time and reagents working on the wrong cells," he says. "At the right price, every lab will adopt this.

People attend a ceremony in honour of the victims and survivors of Lamia flight 2933 on the first anniversary of the plane crash in Colombia that wiped out Brazilian football club Chapecoense, at the Arena Conda stadium, in Chapeco, Santa Catarina, Brazil on November 28, 2017.


The plane was flying Chapecoense to Medellin to take on Atletico Nacional in the Copa Sudamericana finals — the biggest and most unexpected game in the Brazilian team’s history.


When the plane ran out of fuel and went down in inhospitable mountains near its destination, 71 of the 77 aboard died, including 19 players.


Two army helicopters scattered flower petals over a town square Tuesday as part of sad tributes to the upstart Brazilian football team Chapecoense, most of whose players died in a plane crash exactly a year ago.


La Union, the town closest to the site of the accident on a mountainside near Medellin, Colombia, hosted a commemorative ceremony.


The names of the late players and team staff — members of a modest squad that was about to play in a championship game — were read aloud in the town plaza.


The crash killed 71 of the 76 people aboard the plane operated by the Bolivian carrier LaMia. The team lost most of its players and coaches. Only three players survived.

A probe into the crash determined the plane ran out of fuel and was overloaded.


“Glory was close,” said Andres Botero, president of Atletico Nacional, the Colombian team that Chapecoense was going to play in the final of the Copa Sudamericana tournament last year.


“Tragedy crushed their dream,” Botero added.


A memorial plaque was unveiled in the town square as part of the remembrance activities, which were also to feature two Masses and flyover of the crash site by a Colombian air force jet.


A separate memorial ceremony was planned for just after midnight in the town of Chapeco itself.
The plane crash took place a year ago Tuesday Colombia time, early Wednesday Brazil time.


An outpouring of tributes came from towering figures in the world of football.


“Today my heart is in Chapeco,” Brazilian legend Pele wrote on Twitter. “It is a day to remember and pray for those who went to heaven too soon.”


Current Brazilian star Neymar told Spanish TV, “All of us Brazilians carry them in our hearts.”


On the road leading to the hill where the plane crashed — known now as Mount Chapeco — a farmer named Luis Albeiro Valencia has set up a monument erected to those killed in the tragedy.


It is a wooden replica of a plane beside two brick columns. One holds tires from the landing gear of the airliner that went down, the other a deflated football.


He said he was given the items in appreciation for helping with rescue operations that horrible night.


Of the monument, Albeiro Valencia said: “It is to remember. So people will not forget them, because with time everyone will probably forget this hill.”

The popular Nigerian rapper and producer Chibuzo Nelson Azubuike popularly known as the Phyno sank the rumors surrounding a supposed photo of him kissing a guy in a club some time ago, and we did not paraphrase what he said at all. Speaking to the Tribune, he said:

“The only thing that was in my head when I saw the picture was where did this happen. I said back then that a gangstar like me could not have been involved in something like that. I don’t care and I am not bothered. Nigerians have what I call the crab mentality. Have you seen a bunch of crabs in a bucket? When one is trying to come out, the others pull it down. If these insinuations were true, by now there should be a picture to prove it. Look at most successful Nigerian entertainers, they were either tagged gay or Illuminati which I find weird. You see some girls acting their lives away on TV and people tag them as “runs girls”. Almost every young, successful entertainer has a negative tag and it is disheartening.


The word on the streets is that you both live together. Is that true?

Yes we do. But is it anybody’s problem who I live with or who Uti lives with? We all know how expensive it is to live in a highbrow area in Lagos. If you have a friend who youare getting along with and you want to live with each other, is that anybody’s problem?

Yes, it has affected my emotional life, because everybody now believes I am in a relationship with Genny and my family is not happy about it. It has really affected my career and image at home.Have you had any similar issue back home before then?I have never been tagged that before. It is something that is completely new to me. There has never been a time Iwas accused of being a lesbian. One little thinghappened about me and Genevieve Nnaji and everyone now believes I am a lesbian. It is very touching and depressing.


What actually happened between you and Genny that sparked the allegation?
Hmm, you know I speak more of French than English,so, it is a little hard for me to express myself fully. What happened is that Genny and I were in a club in Lagos and her phone got missing. Later, I think a paparazzi or someone got hold of it and started sharing some contents about me and Genny online. I don’t see anything abnormal in the pictures but people started talking that we must be lesbians and all.


What kind of pictures are you talking about?
As normal persons and friends, we do take normal pictures at home and in other places but I don’t know why the online magazine showed Genevieve and I kissing, I am surprised and don’t know where they got those pictures from.


So, you and Genevieve were never kissing each other in any pictures you took together?
“We kissed, but it was nothing extraordinary, it was normal kiss between girls. I mean girls do kiss each other normally, even guys do too. When I was dating Fally Ipupa we used to kiss and now that we are separated and are good friends we still continue kissing like we used to. My fear is I receive strange calls saying that in a few days’ time they will be releasing more pictures about us.”

this are real picture of africa youth suffering in libya and majority of them are from war prone zone like somalia, congo DR, both North and South Sudan, Tanzania, only few from Ghana and Nigeria.

take a trip to the region and come and tell Nigeria what you see there...

I will also employ you to take a trip to Refugee camp in pretoria South Africa where you will see our africa youth suffering and fighting to get an asylum paper to make them reside temporarily in that country, I have make a research to that region before in 2014 and i wept for our africa brother suffering there in SA and i know what i am talking about.
