That's correct, it's like protein engineering but the research field where it's used is what differentiate it. We just developed a technique called Extension Iterative Saturation Mutagenesis (the paper will be published soon). In our research lab, we work with an SH3 protein (a signalling protein). My part of the research: I studied the vital parts of the protein that helps its stability to denaturants and proteases. I chosen one site first, I designed a random saturated mutation (to reduce or optimize it) for the key residue in this site.

This is followed by PCR, transformation into cells, randomly selection and culturing of about 500 cells, protein expression, purification and screening for the best hits by stability assays (using chemical and thermal denaturation). The best hits were screened again until the most stable (if to optimize it) or less stable (if to reduce it) is gotten and the gene for the protein is sequenced to know what's mutated.

Then the gene of the mutated protein is subjected to another rounds of E-ISM and doing this several times on different sites of the same protein, an unstable signalling protein can be optimized and/or elongated or destabilized if reverse is all we need. Also, hormones, peptidomimetics and other transient proteins can be subjected to this technique for enhancement or downgrading.

It's a long and time consuming research.

Thanks. What we're trying to do is modify the winstar rat DNA through it's embryonic stem cell culture. So far we've been able to trigger serious side effect and anomalies due to our choosen method of introducing the modified DNA, we're currently using Zinc Finger Nucleases (ZFN), this method has been proven to work quite well, according to literature review, but we've had no luck. Am sorry if I sound dumb, am just a student research volunteer, trying to publish a paper or two before the end of my undergraduate studies, I hope I would be as good as you one day. Nizstephen: Currently studying statistics and I hope 2 combine my statistical knowledge in the computer sector (computational statistics) currently thinking of working with softwares(SAS) that are statistical in nature to solve problems in diff aspect of life even in business. Am still thinking of more things to embark on when I graduate, but major areas 4 now are *computational statistics and *information research
