It is estimated to have originated over 1000 years ago in Nigeria, and a genome-wide survey of human variation identified the gene LARGE, biologically linked to Lassa virus infection, as among the strongest signals of natural selection in the Yoruba population of Nigeria. We pursued the hypothesis that Lassa virus is an ancient selective force, driving the rise of genetic resistance, by conducting a genome-wide association study (GWAS) of Lassa fever cases and controls in Nigeria and Sierra Leone. We report a signal of association at LARGE, and show that it overlaps with the previously reported signal of selection. This suggests recent evolution of resistance to Lassa fever in West Africa, making it one of only a handful of known loci under positive selection elucidated to be involved in resistance to infectious disease. Population genetics analysis provides further insights into the origins and spread of the resistance variant in LARGE. We show that the same putatively protective haplotype under selection in the Yoruba is also present in the Esan population of Nigeria and the Mende population of Sierra Leone. We find that the frequency is highest in the Yoruba (30%), lower in the Esan (21%) and the Mende (10%), and absent in East African and non-African populations (where Lassa fever is also absent). This is consistent with results from our sequencing of the Lassa genome showing that the virus is oldest in Nigeria and appeared only recently in Sierra Leone, and helps to explain higher fatality rates and greater disease severity in Sierra Leone. Beyond LARGE, our other top GWAS signals fall at genes involved in the immune system, including PTPRE, which modulates expression of cytokines involved in host response to viral infection, and IFRD1, involved in the activation of neutrophils in innate immune response. We also examine signals of adaptation in the Esan and Mende populations and discover numerous novel candidate loci under positive selection, providing new insights into human diversity in West Africa. This work is one of only a few surveys of genetic diversity in West Africa, a region underrepresented in GWA studies. It is also the first ever GWAS on a BL-4 agent, and demonstrates a novel genomics approach to understanding the world’s deadliest pathogens. I just found an academic scholarship by Harvard University and it did corroborate the argument by this hypothesis about LARGE and IL21 genes being resistant to the Lassa Virus. scholes0:Err, if it's based on haplotype - then Fulanis should also be immune to the virus. Yorubas and Fulanis have a lot in common based on haplotype. Anyway, it's very plausible. Most Nigerians don't even know that the genetic make-up of all the groups in the country aren't the same and the occurrence of certain haplotype in the different groups varies. Yorubas for example have more in common genetically with the Bamilekes of Cameroon, Tutsis of Rwanda, Mandinka all over West Africa, Fante/Ga/Ewe folks of Ghana etc. than 99% of all the other ethnic groups in Nigeria. While Igbos for example genetically have more in common with a lot of groups in East/Central Africa than Nigeria. Ditto Hausas and the Sahel region of Africa. Etc. Just cos folks have the same phenotype, that doesn't mean that they're genetically the same, though they're all from a larger tree that branched out into different things. There are certain people, especially in East Africa, that are naturally immune to HIV. Regardless, since it's a Nigerian professor (not dissing the guy) who discovered this - I don't think I'll take it seriously. But there's nothing bad in doing further research on the hypothesis. |

Isn't his honeymoon over? Why are folks giving him a pass, despite his lack of achievements since he took over? Then when you look at his travels and the foreign policy of his government - you would see how much of a failure he has been thus far. He travelled to Iran, came back, and decided to get entangled with the shiites, thus making his travel to Iran a wasteful thing. Then decided to join an unnecessary coalition with Saudi Arabia and Qatar to fight an ISIS both countries created and funded. The Syrian war is about to take a new twist after the combined forces of Russia, Syria, Iran, Shiites, and Hezbollah encircled Aleppo a few days ago and there might be a showdown between those on the Russian bloc vs. those on the US/Saudi/Turkey bloc. So since Nigeria is in coalition with the latter - is he also going to send Nigerian troops there at the mercy of Russian bombs and their special forces? Why's Nigeria involved in something that has absolutely nothing to do with Nigeria? |
