Breaking News: The Great Oba of Benin Reportedly joins his ancestors. His Royal Majesty Omo n’Oba n’Edo Uku Akpolokpolo Erediauwa, the monarch of Benin Kingdom has joined his ancestors. He was 92 years old.According to authoritative sources within and outside Edo State, the well-regarded king transited to the great beyond about two weeks ago.

His death is still being kept secret because by Benin tradition, the death of an Oba could only be officially announced 30 days after his transition.The Oba was said to have died in his palace. Immediately he was confirmed dead by his personal physician, a delegation of senior Benin chiefs went to the house of the heir apparent the throne, Prince Eheneden Erediauwa to inform him of the development.

Investigations by ionigeria news revealed that the Oba died about two days before the campaign rally of the Presidential candidate of the All Progressive Congress (APC), Major General Mohammadu Buhari in Benin City. Buhari had his rally in Benin City on Thursday, January 29, 2015. That is why the Monarch could not receive Major General Buhari.

Similarly, when President Goodluck Jonathan visited Benin City for his campaign rally, he was only received at the palace by a team of Benin chiefs led by the Iyase, Chief Sam Igbe. Oba Erediauwa was born in 1923, son of Oba Akenzua II.

Before being crowned he was known as Prince Solomon, Aiseokhuoba, Igbinoghodua Akenzua. He attended Edo College, Benin City, Government College, Ibadan then Yaba College, before going to King’s College, Cambridge to study Law and Administration. He joined the Eastern Nigeria Civil Service in 1957 as a District Officer, later moving to the Federal Civil Service where he retired as Permanent Secretary, Ministry of Health in 1973.

For a short period he was the regional representative of Gulf Oil. In 1975 he was appointed Commissioner for Finance in Bendel State during the Military Administration of Major-General George Agbazika Innih.Ascending to the throne as the 38th Oba of Benin on 23 March 1979.

During this period, he several times acted as a peacemaker between politicians. For example, he intervened in a dispute between Abia State governor Orji Uzor Kalu and Tony Anenih, Chairman of the People’s Democratic Party Board of Trustees, and resolved another face-off involving Anenih and former Edo State Governor Lucky Igbinedion.

More recently, he tried to broker peace between the feuding members of Edo State House of Assembly.It would be recalled that his wife, Queen Esther died about a year ago. we made some calls to one or two family members, but they neither refuted nor confirm the story to be true or false.

We therefore believe this has a lot to do with the tradition of keeping quiet for 30 days. so we advise you handle this as speculation until the palace releases its official statement, so we do not offend the traditions of the great Bini kingdom.May his soul rest in perfect peace.

In their time we had a sitting governor kidnapped by a thug working for the president. We had a whole region of the country rigged in favor of his party. Morris Ibekwe, a wanted international fraud star won election under PDP.

Those who stole Nigerians blind under PDP; Atiku, Obasanjo, Baraje, Saraki, Wammako, Kwankwaso, Amaechi, Oyinlola, Segun Oni, Okorocha, Ngige, Adeleke, El Rufai, etc, are today the same people financing APC and calling for "CHANGE".

Flashback to 1999, who were these people? Billionaires ? They where nobody. Today, they spend billions and no one dare to ask a question. If you do you are enemy of progress.

They started with "New PDP" when that didn't fly, they joined a cult group called ACN to become APC. Same character with different name.

A very big thank you for all the contributions.Like I said my wife was aware before I spent the money, sometimes, we go to site together, I always carry her along in whatever I do. The problem I have with this is not the money, but not trying to sacrifice for the sake of Family, that is my issue. The reason why I mentioned the house rent thing is this, I expected my wife to think and say, oh! We are paying our rent this month, this will not be the best of time to ask for money, also like I said, if my wife had come to me to ask for money without making reference to the 100000 naira, I would have given her twice that amount.I am saying she should stop giving me the impression that she will not sacrifice for the Family.

Amsterdam, The Netherlands, Thursday 25 April 2013: Exciting new data presented today at the International Liver Congress™ 2013 include results from early in vitro and in vivo studies targeting covalently closed circular DNA (cccDNA), which may form the basis of a cure for chronic hepatitis B virus (HBV) infection.

HBV cccDNA is organized into mini-chromosomes within the nucleus of infected cells by histone and non-histone proteins. Despite the availability of efficient therapies against HBV, long-term persistence of cccDNA necessitates life-long treatments to suppress the virus. The following three experimental studies demonstrate effective HBV-cccDNA targeting/depletion using novel therapeutic approaches which offer the potential of a cure.

Liver regeneration induces strong reduction of viral replication and cccDNA levels, but not complete cccDNA eradication; without antiviral treatment, de novo HBV infection can be re-established

Key findings of research[1] in HBV-infected human hepatocytes using the uPA/SCID chimeric mouse system show that liver regeneration induces strong reduction of viral replication and cccDNA levels, with rapid formation of cccDNA-free hepatocytes. However, because complete cccDNA eradication is not achieved, in the absence of antiviral treatment, de novo HBV infection could be re-established in quiescent (non-dividing) human hepatocytes. This suggests that induction of hepatocyte turn-over together with antiviral drugs inducing viral suppression, such as nucleoside analogues and IFN, or blocking cell entry, may accelerate the clearance of the viral minichromosome.

Targeting epigenetic control of nuclear cccDNA minichromosome to suppress HBV transcription and replication may form basis for other therapeutic approaches to curing chronic HBV infection.

In the infected liver cell the rate of replication of HBV is regulated by the acetylation or methylation of histone proteins which surround the cccDNA minichromosome – so called epigenetic regulation. In a separate innovative study[2], the suppression of HBV transcription and replication by small molecules that target the epigenetic control of nuclear cccDNA minichromosome was investigated. The different classes of small molecules studied included: Class I, II and III histone deacetylase inhibitors (HDACi); p300 and PCAF histone acetyltransferases (HAT) inhibitors; hSirt1 activators; JMJD3 histone demethylase inhibitors.

The combined inhibition of p300 and PCAF HATs resulted in an evident reduction of HBV replication which mirrored the decrease of pgRNA transcription. The hSirt1/2 activator MC2791 and the JMJD3 inhibitor MC3119, albeit with different efficiency, inhibited both HBV replication and cccDNA transcription. Results represent a proof of concept that activation of hSirt1 and Ezh2 (through the inhibition of its functional antagonist JMJD3) by small molecules can induce an active epigenetic suppression of HBV cccDNA minichromosome similar to that observed with IFNα, and lead to persistent cccDNA silencing.

Lymphtoxin beta receptor (LTbR) agonisation represents basis for novel alternative therapeutic approach to curing chronic HBV infection

The final study[3] demonstrated that stimulating the lymphtoxin beta receptor (LTbR) provides an effective, long lasting and non-cytopathic mechanism for achieving effective HBV-cccDNA depletion in infected hepatocytes. Cell culture models including HBV-infected HepaRG cells and primary human hepatocytes were used to test the effect of antibodies stimulating human LTbR (BS1 or CBE11). Results show that a strong and dose-dependent anti-HBV effect was achieved by activation of the LTbR. All HBV replication markers were decreased with this treatment, including cccDNA in cells where HBV infection was already established.

Hepatitis B is the most prevalent cause of chronic viral hepatitis and a major global health problem. Prof. Fabien Zoulim, EASL Educational Councillor commented on the exciting new data: “In chronic hepatitis B infection, the viral genome forms a stable minichromosome - the covalently closed circular DNA (cccDNA) - which can persist throughout the lifespan of the hepatocyte.”

“Current treatments focus on suppression of HBV and discovery of compounds directly targeting cccDNA has been one of the major challenges to curing HBV infection; but these preliminary data show novel therapeutic approaches can be applied to successfully target cccDNA with the long-term aspiration of finding a cure” added Prof. Fabien Zoulim.

The final study[3] demonstrated that stimulating the lymphtoxin beta receptor (LTbR) provides an effective, long lasting and non-cytopathic mechanism for achieving effective HBV-cccDNA depletion in infected hepatocytes. Cell culture models including HBV-infected HepaRG cells and primary human hepatocytes were used to test the effect of antibodies stimulating human LTbR (BS1 or CBE11). Results show that a strong and dose-dependent anti-HBV effect was achieved by activation of the LTbR. All HBV replication markers were decreased with this treatment, including cccDNA in cells where HBV infection was already established.

Hepatitis B is the most prevalent cause of chronic viral hepatitis and a major global health problem. Prof. Fabien Zoulim, EASL Educational Councillor commented on the exciting new data: “In chronic hepatitis B infection, the viral genome forms a stable minichromosome - the covalently closed circular DNA (cccDNA) - which can persist throughout the lifespan of the hepatocyte.”

“Current treatments focus on suppression of HBV and discovery of compounds directly targeting cccDNA has been one of the major challenges to curing HBV infection; but these preliminary data show novel therapeutic approaches can be applied to successfully target cccDNA with the long-term aspiration of finding a cure” added Prof. Fabien Zoulim.

[/b]Novel Therapeutic Approches to Cure Chronic HBV Infection
Amsterdam, The Netherlands, Thursday 25 April 2013: Exciting new data presented today at the International Liver Congress™ 2013 include results from early in vitro and in vivo studies targeting covalently closed circular DNA (cccDNA), which may form the basis of a cure for chronic hepatitis B virus (HBV) infection.

HBV cccDNA is organized into mini-chromosomes within the nucleus of infected cells by histone and non-histone proteins. Despite the availability of efficient therapies against HBV, long-term persistence of cccDNA necessitates life-long treatments to suppress the virus. The following three experimental studies demonstrate effective HBV-cccDNA targeting/depletion using novel therapeutic approaches which offer the potential of a cure.

Liver regeneration induces strong reduction of viral replication and cccDNA levels, but not complete cccDNA eradication; without antiviral treatment, de novo HBV infection can be re-established

Key findings of research[1] in HBV-infected human hepatocytes using the uPA/SCID chimeric mouse system show that liver regeneration induces strong reduction of viral replication and cccDNA levels, with rapid formation of cccDNA-free hepatocytes. However, because complete cccDNA eradication is not achieved, in the absence of antiviral treatment, de novo HBV infection could be re-established in quiescent (non-dividing) human hepatocytes. This suggests that induction of hepatocyte turn-over together with antiviral drugs inducing viral suppression, such as nucleoside analogues and IFN, or blocking cell entry, may accelerate the clearance of the viral minichromosome.

Targeting epigenetic control of nuclear cccDNA minichromosome to suppress HBV transcription and replication may form basis for other therapeutic approaches to curing chronic HBV infection.

In the infected liver cell the rate of replication of HBV is regulated by the acetylation or methylation of histone proteins which surround the cccDNA minichromosome – so called epigenetic regulation. In a separate innovative study[2], the suppression of HBV transcription and replication by small molecules that target the epigenetic control of nuclear cccDNA minichromosome was investigated. The different classes of small molecules studied included: Class I, II and III histone deacetylase inhibitors (HDACi); p300 and PCAF histone acetyltransferases (HAT) inhibitors; hSirt1 activators; JMJD3 histone demethylase inhibitors.

The combined inhibition of p300 and PCAF HATs resulted in an evident reduction of HBV replication which mirrored the decrease of pgRNA transcription. The hSirt1/2 activator MC2791 and the JMJD3 inhibitor MC3119, albeit with different efficiency, inhibited both HBV replication and cccDNA transcription. Results represent a proof of concept that activation of hSirt1 and Ezh2 (through the inhibition of its functional antagonist JMJD3) by small molecules can induce an active epigenetic suppression of HBV cccDNA minichromosome similar to that observed with IFNα, and lead to persistent cccDNA silencing.

But new research coming out of the Texas Biomedical Research Institute at San Antonio indicates that a Hep B cure could be on the way.A drug developed by Gilead Sciences has proven itself to be able to destroy cells infected with the hepatitis B virus (HBV) in chimpanzees that have the chronic form of hep B. Chimps are the only other primate besides humans that is able to contract a chronic form of hepatitis.The drug is called GS-9620 and is an immune system stimulator. It works with the body’s natural defense system to fight off HBV, eliminate infected cells and reduce the number of infected cells in the liver of the chimpanzees. The success at Texas Biomed with the primates was critical in allowing it to move forward into the human trial phase.A full report of the study co-authored by research scientists from both Texas Biomed and Gilead Sciences can be found in the May issue of Gastroenterology magazine.The drug is not yet a cure, but it promises to be a stepping stone on the way to completely eradicating the disease. The study claims that similar viruses to HBV in other animals can be cured using GS-9620 treatment. The fact that this eliminative potential exists in the drug at all shows that through some minor adjustment, it is possible that aggressive treatments of GS-9620 derivatives could ultimately lead to total dissolution of hepatitis B infections.“This GS-9620 therapy represents the first conceptually new treatment for HBV in more than a decade, and combining it with the existing antiviral therapy could be transformative in dealing with this disease,” said Robert E. Lanford, Ph.D., co-author of the study and researcher at Texas Biomed.

I am a christain, and i believe in God and that christ died for me. Some time ago, out of desperation, i reduced my age by 2years in other to get a job, i finally got the job and i am doing fine on the job, though later on i asked God for forgiveness, but each time i receive my salary, something keep reminding me that i am paying TITHE from the proceeds of a fraudulent act. Please I need reasonable and a man of God's opinion .
